I have the same on my boat trailer, after cleaning the bearings and putting the seals on correctly, the best way to fill the cavity is with a grease gun, don't for get the bearing buddies, they have a habit of falling off.

The bearing buddies have a grease nipple which is used to keep the grease up to the bearings, depending on the usage, if frequent, it is perhaps best to at least clean and change the grease at least once a year or more frequently if the bearings get a regular dunking in (salt) water.
